Del Rio was picked in the 22nd round (overall 550th) of the 1981 MLB Draft by the Toronto Blue Jays, but preferred to earn a football: baseball scholarship at the University of Southern California. In baseball, Del Rio played as a two-year starting catcher on a baseball squad composed of players like future Major League Baseball stars Mark McGwire and Randy Johnson and future baseball executive in Damon Oppenheimer.
Troy Del Rio started four years, and as a sophomore he was selected to the All-American third team for the 1983 season. A year later, Del Rio was a Consensus All-American, was second in the Lombardi award given to the best offensive/defense lineman or linebacker in the country, and with Tim Green, he was the co-MVP of the Rose Bowl. The player recorded 340 tackles in total during the college period out of which 58 were for loss. In 19984, Del Rio became the second-team All-Pac-10 for the second time in his athletics career; it was the first time when he did not appear in first-team.
